EXAKT Thin Section Cutting Technology 
Perfect sample preparation

In medicine, the high-precision EXAKT thin section cutting system is used for the preparation of histological preparations, so-called thin sections, with a thickness down to 10 µm. Bone samples need not be decalcified for this. Thus they retain their cell information and morphology. The method of Thin Section Cutting Technology is especially important in the study of interfaces of implant/bone, bone/cartilage etc. since the interfaces can be prepared while preserving the structure and used directly for light microscopy techniques.

Among other fields, the system is used in the research and development of implants, stents, cardiac catheters, in oral pathology, dentistry, orthopedics, veterinary medicine or immunohistochemistry. It was developed in collaboration with Prof. Donath, Institute for Pathology at the University of Hamburg in 1987 and since then has proven itself to be unparalleled.

Gently cutting the uncuttable 

The high-precision EXAKT 300 and 310 diamond bandsaws are used to cut highly sensitive tissue and tissue combinations for the histological analysis of bone and muscle tissue, in particular bone with implants or screws. Fresh as well as embedded samples can be processed.

The crucial interface between the different tissue structures is optimally prepared. Burr-free, plane-parallel surfaces and thicknesses of down to 100 µm allow many slices to be cut in order to obtain as much structural information as possible from the original sample. 

A very wide range of sample sizes can be processed – from dental clinical centers or cardiology to veterinary medicine, for instance.

To adapt the process to the material and workflow, the EXAKT system provides alternative cutting characteristics that are distinguished by the contact of the sample to the cutting belt, CL (Contact Line) and CP (Contact Point). Many different shapes can be accurately processed with a variety of clamping options.

Precise, safe and simple for everyone

The current conventional processing of large samples with a butcher's saw has so far produced only very poor results that are not very meaningful and inadequate for diagnostics.

Handling was also extremely dangerous.

The EXAKT 312 allows quick, precise preparation of the native condition of a sample in order to best support further processing, such as fixing, dehydration and decalcifying, and hence create the precondition for meaningful diagnostics.

Furthermore, the entire handling is very safe. Tissue is not sawn, but ground very finely with the diamond cutting band. This massively reduces the risk of an accident. The EXAKT 312 also complies with the latest standards for safety and hygiene in medicine, which ensures faster and more efficient workflow.

MICRO GRINDER
EXAKT 400 CS 

Perfect and micron-precise

The 400 CS micro grinder is an elementary component of the preparation process of a sample for constant plane-parallel thin grinding down to 20 µm with excellent surface quality.

The high-precision engineering is based on a unique machine concept with which all relevant information, e.g. the interfaces between bone and implant, adhesions, tumors or penetration in new and old bone can be optimally obtained.

Electronic control of the grinding process ensures constant precise results down to an accuracy of µm. After completion of the grinding process, the thin sections are dyed with processes such as Toluidine blue, Masson-Goldner, Van Gieson or Giemsa.

The final grinding pattern differs substantially in quality and informative value from samples that are cut in the conventional manner.

DEHYDRATION AND INFILTRATION SYSTEM

Optimum penetration

The accurate dehydration and infiltration of a sample with plastic resin, such as TECHNOVIT, is an elementary step in sample preparation. Insufficient infiltration is connected with loss of information and can lead to the loss of the valuable sample. For optimum infiltration, up to 6 diffusion stages can be processed at the same time under light-protected conditions. A gentle shaking movement of the unit ensures optimum dehydration and penetration of the resin. If required the final step can also be under vacuum to eliminate any risk of artifacts.

Embedding with TECHNOVIT 7200

Not only hard, but gentle – many factors play a role in the preservation of tissue when embedding histological samples in plastic resin.

To avoid blistering and cracking, the curing time, degree of curing and the temperature development must be controlled and adapted to the material. Based on the TECHNOVIT 7200 plastic from HERAEUS KULZER, EXAKT has developed the 520 light polymerization unit that fulfills these requirements.

Precise and plane-parallel

To process thin sections down to 10 µm, the embedded sample must be mounted absolutely precisely and plane-parallel on a slide. The slide is held by vacuum in order to optimally align and fix the sample. The gluing surface of the embedded sample does not need to be prepared beforehand.

Perfectly bonded

The slide mounted embedded sample must then be bonded exactly plane-parallel  to a second slide for the optimum preparation of a thin section. The bond must be uniform, transparent and free of air bubbles, and the layer thickness of the bond must be measurable. This is achieved by the EXAKT 402 precision adhesive press together with the light-cured adhesive TECHNOVIT 7210 from HERAEUS KULZER.

BLOCK DRYING AND POST-INFILTRATION 
EXAKT 530

Reworking samples

Is there still liquid in the sample? Was it insufficiently infiltrated or have bubbles occurred? These and similar questions come up at the end of a thin section preparation. Valuable samples can be »rescued« and the information content secured with the EXAKT 530 block drying and infiltration unit. Thus, for example, through vacuum and controlled heat, residual water can be drawn from an insufficiently de-hydrated sample, resin added and cured under blue light.
