Teamwork! EXAKT - devices for expanded sample preparation

The device line we offer arose thanks to the parallel development of the process, the embedding plastics, and our devices. All components are optimally attuned to one another with respect to their application and effectiveness. This only came about through close cooperation with Prof. Dr. Dr. Dr. Karl Donath, UKE Hamburg, Germany and Heraeus Kulzer, Germany.

An embedding medium that hardens without exothermic effect and only with blue and yellow light (450 nanometer) was developed. It distinguishes itself through low shrinkage and its excellent cutting and grinding characteristics. A Clogging of the tools is prevented. We offer various embedding media and various processes depending on the application.

EXAKT 510 – de-hydration and infiltration device

 

This ssystem was developed for de-hydration and infiltrating samples with plastics (for example, Technovit 7200), for application in medical and biomaterial research.

With increasing development of cut, thin-ground technology, the problems of preparing bones that are not decalcified with and without implants also increased. Open questions such as remaining water in the tissue and insufficient infiltration drove the device development to advance. 

The E 510 is tailored to users with small numbers of samples but high quality demands. Essential for the diffusion procedures is the change of the media concentration on the tissue surface. This means that the liquid leaving the tissue does not remain on the surface, but it is rinsed away through permanent agitation. 

For a de-hydration and infiltration program of 12 days (standard), with its six light-proof containers, the E 510 offers the opportunity to take another step with increasing alcohol concentration every two days. In routine operation, after 12 days a container with the finished prepared samples can be removed for the first time, and every two days, new samples are available.

Samples with bone cement on GMA basis can be de-hydrated under vacuum.

EXAKT 520 – light polymerization device

The EXAKT 520 light polymerization device is used to embed medical and industrial samples in plastics.

EXAKT 530 – re-infiltrator

After cutting open a polymerized block, sometimes tissue zones appear that are not sufficiently infiltrated and polymerized. The most frequent cause of this is insufficient de-hydration. If tissue zones are still soft, the sample is difficult to cut and grind, it clogs up the tools. If, however, the sample is processed further regardless, there is a danger that during the grinding and polishing, water can get into the flaws and detach the sample from the object carrier in the last phase of the thin-grinding procedure.

With the EXAKT re-infiltrator, the poorly-infiltrated preparation can be dried, re-infiltrated with liquid plastic (Technovit 7200), and then polymerized sufficiently.

For users who work with bones or with thick samples that are embedded in plastic, the selection of our re-infiltration process is the variant that saves time and money.

EXAKT 401 – vacuum adhesive press

For histological samples and for other applications that require thin grinding (10 - 25 µm), it is absolutely necessary to work plane-parallel during the entire sample processing. The EXAKT 401 vacuum adhesive press was developed in order to apply plane-parallel samples to slides without having to grind the surface in advance. The vacuum adhesive press works, for example, in combination with Technovit 4000 adhesive.

EXAKT 402 – precision adhesive press

If you want to process a tissue with an implant into a thin-ground sample, the key to success is the perfect placement of the sample on the grinding medium. For transmission microscopy, the transparency of the adhesive layer in combination with the object carrier is just as important.

For the further optimization of the cutting/ thin-ground process, an adhesive (Technovit 7210) was developed in cooperation with Heraeus Kulzer. This adhesive is clear, it enables a parallel adhesive coating, and it hardens only with the application of blue light.

The transparent support bracket of the vacuum adhesive press enables the researcher and the technician to observe the distribution of the adhesive and detect possible air incursions immediately. The necessary corrections can be made easily before the polymerization.

The use of the precision adhesive press enables the developer and the technician to create perfect, even thickness adhesive layers.

Consumables - embedding media and adhesives

EXAKT Technovit 7200 VLC embedding plastic

EXAKT Technovit 7200 is the first light-hardening embedding plastic for tissue for application in combination with the EXAKT cut thin-ground technology. It consists of only one component and is suited for embedding both hard and soft tissue. Under the influence of light, it polymerizes at a temperature of less than 40°C. It is compatible with most water-soluble histological dyes.

EXAKT Technovit 9100 new embedding plastic

Immuno and enzyme-histochemical dyes with samples embedded in plastic are now possible with the new embedding plastic Technovit 9100 new based on MMA. The samples are cut with an EXAKT band saw and then ground on an EXAKT grinding system.

EXAKT Technovit 7230 VLC adhesive

The EXAKT Technovit 7230 adhesive is used to fix samples on the bottom of embedding moulds before they are filled up with EXAKT Technovit 7200 embedding plastic. The position of the later cut surface can be influenced this way.

EXAKT Technovit 7210 VLC adhesive

EXAKT Technovit 7210 is a light-hardening transparent adhesive used in combination with the EXAKT 402 precision adhesive press. It permits optimal positioning of the sample on the slide and, in combination with the precision adhesive press, guarantees the greatest plane-parallelism.

EXAKT Technovit 4000 adhesive

The EXAKT Technovit 4000 is used in combination with the EXAKT 401 vacuum adhesive press to glue sample blocks. As a high-viscosity adhesive, it compensates for all unevenness in the sample block due to the polymerization.

Plastic Slide

The plastic slides offered by EXAKT provide with the greatest plane-parallelism. They are perfectly suited for use in combination with the EXAKT cutting and grinding devices. We offer them in various sizes and strengths.

Embedding moulds

The EXAKT embedding moulds are available in various sizes and fit into the EXAKT 520 light polymerization device.
